V posledních desetiletích se u nás stejně jako v celém světě zvyšuje počet případů degenerativního onemocnění centrálního nervového systému, které se nazývá Alzheimerova choroba. Toto onemocnění u nás postihuje asi 130 tisíc lidí a ve Spojených státech amerických je počet odhadován na 4 milióny. Objevuje se nejčastěji po 65. roce věku, kdy je prevalence 10%, ale může se vyskytnout i u mladších jedinců. Jde o primárně degenerativní onemocnění se zřetelnou genetickou složkou, jehož jednou z hlavních charakteristik je centrální cholinergní insuficience. Průběh Alzheimerovy choroby se vyznačuje především postižením vyšší nervové činnosti ve smyslu postupného úbytku kognitivních funkcí, narůstajících behaviorálních potíží s následným snížením soběstačnosti. Nejdůležitějším rysem je progredující povaha onemocnění s postupující demencí. Prevalence choroby stoupá s věkem a u osob nad 85 let dosahuje až 50%.
