Zavedení antipsychotik do klinické praxe více jak před čtyřiceti lety bylo velkým přínosem pro nemocné, ale i pro jejich lékaře, kteří dostali do ruky další možnost, jak zmírnit utrpení svých duševně nemocných pacientů. V současné době patří tato skupina psychofarmak k nejužívanějším lékům v medicíně. Od použití a zavedení chlorpromazinu do klinické praxe se mnohé změnilo. Psychiatr nyní požaduje od této lékové skupiny příznivé komplexní ovlivnění všech psychotických příznaků včetně pozitivního ovlivnění kognitivních funkcí a spolu s neurologem a internistou co nejmenší přítomnost vedlejších nežádoucích účinků. Na základě těchto požadavků se jeví jako velmi perspektivní nová tzv. atypická antipsychotika (APS). Z nich pak zejména skupina multireceptorových antagonistů- MARTA (Multi - Acting Receptor Targeted Antipsychotics): clozapin, olanzapin, quetiapin. Clozapin je vzhledem ke svému možnému myelotoxickému účinku vyhrazen pouze pro skupinu farmakorezistentních nemocných.
